The thing is that Saitama doesn't have Uber Strenght.

He has Uber Omni Plot Convenience. He is as a strong as the jokes demands him to be.
The joke is that he beats people with one punch. So he wins.

Thanos will only win if he can get the first shot in, he has to destroy Saitama before he can unleash a single punch. Can he do it? I don’t know, but it’s the only way to keep from becoming another exploding head.

Pfft. Nah. That'd be like Elmer Fudd actually shooting Bugs Bunny in the head and killing him. There's a reason you never see that. That punch would HAVE to pulp Thanos, just BECAUSE it's Saitama giving it.

Saitama operates on cartoon logic, he can never lose. Yet at the same time, it's why he always feels so blank and empty. He craves a challenge and the thrill of a real fight, so by winning every time, he actually loses every time. He'd welcome an opponent who could take him out, that's why he'll never find one. The only tension in One Punch Man is generated by the other heroes, who don't have Saitama's double edged plot armor.

Truthfully, Thanos is mulch, and Saitama goes back to his hovel to wash his gloves in disappointment.

Doesn't matter. If he feels like it, Caped Baldy wins with one Serious Series: Serious Punch. Can't be the One Punch Man if there's anything he can't one-punch, you know?

This encounter entirely depends on if A) Thanos or Saitama knows what eachother can do. and B) Thanos has the Infinity Gauntlet.

If Thanos knows that Saitama is undefeated, and can (for the most part) defeat his opponents in one punch, all Thanos would need to do is snap his fingers and either remove Saitama from reality, or completely nerf him into the ground before pummeling him.

This is of course taking into account that Thanos has the Infinity Gauntlet.
Without it, the tables would turn quickly in favor of Saitama rather. But then you also have to keep in mind, Thanos was effectively banned from entering the realm of Mistress Death (Whether this is still the case, i don't know), making him truly immortal.

If Saitama knows what Thanos can do with the Infinity Gauntlet, and isn't going to mess around with him just for a good fight? All Saitama would need to do is get in close and punch him once and
hopefully end it there.
HOWEVER... Saitama has had battles in the past that have lasted "beyond one punch." His final battle against Boros in the first season being a perfect example for that (I've only seen the anime. So that's where my knowledge comes from).

So in the event that Thanos has the Infinity Gauntlet, the fight lasts longer than the initial punch from Saitama, and Thanos is beginning to lose. All he'd have to do is snap his fingers and wipe Saitama out of reality itself. Thus winning the encounter handily.
Saitama's plot device of "always winning" wouldn't matter at that point if he didn't exist in the first place.

Without the gauntlet, Saitama wins. If not from killing Thanos outright (which he can't actually do because: Immortal), or knocking Thanos into the next galaxy, which would really only serve to delay another fight.

Considering Thanos without the gauntlet is at least on par with Boros, if not higher, it makes for an interesting fight. I believe  that Saitama holds the edge against Thanos without it, but with it I won't matter what OPM does.

Thats a fairly good question, thats kinda slanted in OPM's favor. Marvel has a character with the exact same premise of OPM, and was made for the exact same reasons as OPM. Squirrelgirl. And though it was just a "clone" of thanos, people who should have known it wasnt the real thanos was still so convinced that it was him (like the watcher), and she already beat him easily.

Though that "clone" didnt have an item that could reshape and/or erase existence with him... OPM's sole purpose is to be ridiculously overpowered and win everything.

But unless OPM is able to break the 4th wall and tell comic creators what to do, if anyone/anything would be able to defeat him, it would be an item which's purpose is to essentially be the writer's & artist's pen

Squirrel Girl is actually a gag character. Saitama is a parody character. All the people he beats are featless with the exception of Boros and Garou. Squirrel Girl is like the inverse. She could beat Living Tribunal and you would buy it. She scales off their feats. Saitama and his enemies are like a black hole of feats. So you can't scale shit. He, at best, beat a planet buster. That's it. That doesn't mean he can solo Marvel. COME ON. You know how many planet busters Marvel and DC has? LOTS.
